We've done this boss multiple ways. You can 2-tank+RangeTank and 3-heal it if you want, it will just take a little longer but you shouldn't have to worry about enrage timers.
Now days our 10 man has one tank do both of the melee guys and the other tank does the ranged guy, with 2 healers. In the past we have the two tanks do the two melee guys and a caster (we've used Warlock and Shadow Priest to good effect) take Keleseth. We've done it this second way with both 2 healers and 3 healers. So you have a lot of choices.
To answer your question, you should have no problems tanking Keleseth, and you don't need special gear, buffs, or spec to do it. The DoT damage from having Nuclei on you is pretty negligible, so as long as you have at least 4 Nuclei on you before Keleseth gets empowered you'll be fine. Our Shadow Priest was running around with 5 or 6 on him sometimes. Maybe if you have some PvP gear (i.e. some extra Stamina) that can be a little buffer for your healers.
You have two hunters, so make sure they take the time to Misdirect Keleseth to you whenever they can so you can get some decent threat going. Spam some of your own spells into Keleseth, then run around and toss some Ice Lances at every Nuclei you see. And make sure not to outrange your healers...

okay thank you, but will resistance lower the damage that you get?
Yeah, I would certainly run with Shadow Protection (since you have a priest to cast it), but I wouldn't bother trying to find, gem, or enchant any gear for Shadow Resistance. The big key is to make sure you have enough Nuclei on you and you'll be fine.
