I don't think its possible to see the cooldown of Mortal Strike from a macro like this. Macros, as far as i know, only show the component that has the longest cooldown. You could try to have MS on an action bar in the corner of your screen or something to show the cooldown on that.
For the structure of the macro, you're looking at something like this (basic one here, others can probably tell you a neater way of putting it):
/use <trinket name>
/cast Mortal Strike

Some of the more useful ones are on one of the stickies not sure which one exactly. Among them are the macro for spell interrupts, and the correct charge move to use based on target and whether or not you're in combat.

For clarity, combing what Malekith and Huin said will get the macro you wanted, with MS cooldown:#showtooltip Mortal Strike
/use
trinketnamehere
/cast Mortal Strike

Macros can only trigger one Spell/attack that initiates the GCD, per click. That is a hard limit Blizz put on macros to stop "Botting". "Using" a trinket does not, as far as I can tell, trigger the GCD. So you should be able to do what you want with one click.
